What is PT-141?

PT-141 (Bremelanotide) is a cyclic heptapeptide melanocortin receptor agonist derived from Melanotan II. It was FDA-approved in 2019 as Vyleesi for premenopausal women with hypoactive sexual desire disorder (HSDD) โ€” making it the only peptide specifically approved for sexual dysfunction.

Unlike Viagra and Cialis, which work peripherally by dilating blood vessels, PT-141 acts centrally through the nervous system โ€” specifically activating MC3 and MC4 melanocortin receptors in brain pathways associated with sexual arousal. This central mechanism means it works through desire and arousal rather than physical blood flow, and is effective in cases where PDE5 inhibitors are not.

Key Benefits

โœ“FDA-approved mechanism (Vyleesi)
โœ“Increases sexual desire and arousal
โœ“Works centrally โ€” not dependent on physical stimulation
โœ“Effective for both men and women
โœ“Fast onset via sublingual melt delivery
โœ“No dependency or hormonal effects

How It Works

PT-141 activates melanocortin receptors in the hypothalamus and limbic system โ€” areas of the brain associated with sexual arousal and motivation. Activation of these receptors increases dopamine signaling in reward circuits, producing increased sexual desire and arousal.

This mechanism is fundamentally different from PDE5 inhibitors: PT-141 works whether or not there is physical stimulation and does not depend on nitric oxide signaling or vascular response. It addresses desire at the neurological level.

Research Highlights

Phase 2 and Phase 3 clinical trials leading to FDA approval documented significant improvements in sexual desire, arousal, and satisfaction in women with HSDD. Studies in men show improvements in spontaneous erections and sexual satisfaction. The combination of PT-141 with oxytocin has been studied for additive effects on both arousal and emotional connection.

Subling delivery of PT-141 bypasses first-pass metabolism and provides faster onset than oral administration. Our melt strip format is designed to match the onset profile of the clinical delivery routes used in research.

Frequently Asked Questions

What is PT-141?+
PT-141 (Bremelanotide) is a cyclic peptide that was FDA-approved in 2019 as Vyleesi for sexual dysfunction. It works by activating melanocortin receptors in the brain that are involved in sexual arousal and desire.
How is PT-141 different from Viagra?+
Viagra (sildenafil) works peripherally by dilating blood vessels. PT-141 works centrally through the nervous system โ€” increasing desire and arousal at the brain level. They target completely different mechanisms, which is why PT-141 can be effective when PDE5 inhibitors are not.
How fast does a PT-141 melt strip work?+
Sublingual delivery typically produces effects within 30โ€“60 minutes. Onset can vary based on individual factors. Most users notice effects within 45โ€“90 minutes.
Can PT-141 be used by both men and women?+
Yes. Clinical research has been conducted in both sexes. FDA approval is specifically for women with HSDD, but research in men documents improvements in sexual function and satisfaction.
